CRISPR / Cas9 method: The cutting edge technique

This simple and fast method of changing genes has revolutionized the scientific world. The CRISPR / Cas9 method allows very fast and specific genome editing.

This technique has already been used successfully in many model organisms (yeast, plant, zebrafish, Drosophila, nematode and mouse) and also works very well in human cell lines. As a result, it has great potential to be used as a therapeutic agent in the near future. You can find more information about this in our information  center.

Genome editing with the help of CRIPR / Cas9 requires two components in particular: an endonuclease and a  guide RNA (gRNA).

abm offers a wide range of Cas9 stably expressing cell lines . These are available for all popular research models. Cell lines with a cumate-inducible system can also control expression and control it over time. All cell lines are pre-validated by qPCR and / or by Western blot  . For successful genome editing, the gRNA must be introduced into the cell lines using viral transduction or transfection. They can then be checked for a gene knockout.

There is also the possibility  of transfecting Cas9 proteins together with the gRNA directly into existing cells in order to edit the genome (available on request for AAV, lentivector, adenovirus and non-viral vector expression systems). This is a very quick method because there is no transcription or translation. The Cas9 proteins offered by abm can be used on many organisms without having to consider codon optimization or promoter compatibility. In addition, this type of CRIPR / Cas9 method does not require plasmids or viruses, and therefore excludes integration into the genome.
